Hedgehog Pathway Inhibitor
Itraconazole is an antifungal, similar to ketoconazole. Ketoconazole has been used to suppress the enzymatic production of androgens in the adrenal glands. Unlike ketoconazole, itraconazole is a potent suppressor of the Hedgehog Signaling Pathway that cancers may use for growth. It is currently finding a use in reversing basal cell carcinoma.
A small study last year compared low dose to high dose itraconazole in men with metastatic PC, and found some activity.
